To address this he conducts the following experiment i) he mixes the mRNA encoding the protein with ribosomes in a cell lysate that contains all the machinery required to support translation as if it were occurring in a cell
ii) he then adds ER derived microsomes to the reaction
iii) he waits for translation to be completed
iv) he then adds proteases to the mixture
v) the purifies his favorite protein out of the mix; he calculates from the DNA sequence that the protein is likely to 500 aa long
34) Assuming that the protein is an ER lumenal protein
