Creating a DNA Fingerprint: You will be creating five different DNA Fingerprints based on the "lab" using restriction enzymes and gel electrophoresis process. Then you will use those DNA Fingerprints to answer the analysis questions and solve the case! The Case: Police. are investigating the tragic murder of a young boy. The police recover DNA from skin cells found under the nails of the boy from where they suspect he scratched the attacker., Police suspect the mother and / or the father. The father claims the attacker was the neighbor, whom we saw in the back yard the night of the attack, and who has been living next door for years and has gotten into fights with the father. He thinks this may be revenge against him. The police obtain a warrant to get DNA samples from the mother, father, and neighbor. The Process: You will cut each of the DNA samples below (only one side of the DNA is given but you can complete the second side using base pairing rules) with a restriction enzyme that attaches to the site GTAC and cuts between the T and the A. Then, you will create the likely gel results on the given gel "box" by adding a band for each DNA piece in that sample. Restriction Enzyme: GT / AC Boy's DNA Sample A: Under Skin Nails Sample B: Sample C Mother: Father Sample D: Sample Neighbor E:

1. Based on the DNA results, who did the boy scratch? Defend your answer to the jury by explaining how DNA can be used for identification and reference specific DNA bands in this test. 2. As a forensic scientist, you note something interesting when you examine the DNA of the boy, the mother, and the "father". Look closely at these three DNA samples. What conclusion can you provide about the likely paternity of the father? Defend your conclusion to the jury by explaining how DNA tests can prove / disprove paternity and reference specific DNA bands in this test. 3. Now examine the DNA in samples A, C, and E carefully. What surprising conclusion can be supported? What motive for the murder might this offer? Defend your answer to the jury using the DNA evidence and specific DNA bands.
