Explanation:

Recombinant DNA technology is the heart of applied molecular biology. Recombinant DNA Technology is used to produce Peptide hormone such as insulin,growth hormone various vaccine such as hepatitis B vaccine.

           The pathogenic property of hepatitis B vaccine is present in its surface antigen or surface protein.The DNA that encodes the surface protein is at first isolated by taking antigenic part of the virus.

           The isolated DNA molecule is clonned in a vector and Recombinant DNA is then cultured within yeast Saccharomyces cerevisiae. The cells are allowed to grow in a fermentor which ultimately result in the formation of surface antigen of hepatitis B virus.

         The surface antigen is then purified by various chemical process and the purified protein is then treated with formaldehyde in phosphate buffer along with alum salt .The formaldehyde is used to disrupt the antigenic property of the virus which leads to detoxify the viral toxins.
